- Abstract
- In this paper, new methods to discover and characterize activity of exoenzymes in surface ecosystem water were developed. Enzyme activity was exemplified by proteolitic activity. The new methodology uses measurement of polarization of fluorescence of proteins labeled with fluoresceinisothiocyanate (FITC). This methodology allows measurement of proteolitic activity of exoenzymes in aquatic environment. Using this approach, enzymatic (proteolitic) activity was discovered in natural samples of water (with the natural community of plankton) from a surface freshwater ecosystem in the central region of the European part of Russia. Proteolitic activity was discovered in the water sample before and after removal of phytoplankton and bacterioplankton by water filtration. This provides evidence for making conclusion on the presence of exoenzymes in water medium.
